I have a igt s2000 5 reel multidenom machine and yesterday I tried to play it and it says testing memory and then the reels spin 1 time and the it repeats testing memory. It worked fine before any suggestions

Welcome! :) I would suggest checking for loose connections within the machine. Another thing to check is if the Green Varta battery is still on the MPU? This battery isn’t required for the machine to operate and it can leak acid which can damage the MPU. I would recommend removing it as soon as you can if possible. I would also check to see if the coin battery which is on the MPU is at 3 or more volts. Sometimes, the machine can throw weird errors if the battery is below 2 volts.

No, the machine when turned on originally said testing memory and the reels spin on time very slow, and now it just says testing memory and doesnt do anything else

Slow reels? Sounds like a tilt. Open the door while they are slow spinning & turn the jackpot key. It should stop the reel tilt & might resume normal play after closing the door. If not look for an error message on the VFD & let us know what it is.

The reels only spun one time very slow. And now nothing but testing memory. I've tried the key when the reels spun but that didn't do anything. Took board out and checked,took reels out and replaced took bill acceptor out and replaced. Was playing fine and then nothing.

Please measure the coin battery voltage and also post a picture of the MPU board (the board in the metal tray)
Also make sure you do not remove any boards etc. with the power on. Only exception would be the reels but I don't even like to do them with the power on.
